Tag antibodies

Tag antibodies are applied to detect and purify the multiple proteins with the same tag. Tag antibodies, also known as epitope tag antibodies, are highly specific monoclonal and polyclonal antibodies for the purification and detection of the tagged proteins. Epitope tags are small peptides with addition to proteins of interest in recombinant DNA techniques. These tags are perfect for the research of recombinant proteins, once protein-specific antibodies are not available or suitable for experimental design.

Epitope Tags

The epitope tag is a biological structure or sequence like a protein, peptide, small molecule or carbohydrate, it acts as an antigen to be recognized by the specific antibody. Epitope tagging is the technology to fuse epitopes to the recombinant protein by genetic engineering or chemical conjugation. The epitope tags are able to detect proteins once no antibody is available. This technology is normally applied to characterize new protein discoveries and low abundant proteins.

Epitope tags are useful tools for several immuno-assays, including western blot, immuno-precipitation, immuno-fluorescence. In addition, it combines with affinity chromatography to purify target proteins from complex samples.

Types of Epitope Tags

As different epitope tags are available, the tag choice is dependent on several factors.

  1. Fluorescent peptide: Fluorescent tags Green Fluorescent Protein (GFP), Allophycocyanin (APC) are used for cell tracking and cellular localization.
  2. Protein purification: MBP, His, V5 and Glutathione S-Transferase (GST) are mainly for protein purification.
  3. Protein analysis: C-my, Hemagluttinin (HA), and DYKDDDDK are used in pull-down/immuno-precipitation experiments for protein-protein interaction verification and identification.
